ABSTRACT

Diffuse intrinsic pontine glioma (DIPG) is the main cause of brain tumor-related death among children. Until now, there is still a lack of effective therapy with prolonged overall survival for this disease. A typical strategy for preclinical cancer research is to find out the molecular differences between tumor tissue and para-tumor normal tissue, in order to identify potential therapeutic targets. Unfortunately, it is impossible to obtain normal tissue for DIPG because of the vital functions of the pons. Here we report the human fetal hindbrain-derived neural progenitor cells (pontine progenitor cells, PPCs) as normal control cells for DIPG. The PPCs not only harbored similar cell biological and molecular signatures as DIPG glioma stem cells, but also had the potential to be immortalized by the DIPG-specific mutation H3K27M in vitro. These findings provide researchers with a candidate normal control and a potential medicine carrier for preclinical research on DIPG.

ABSTRACT

Objective To study the value of new 16S rRNA gene chip in pathogen detection of neonatal sepsis.Method Newborns with suspected sepsis hospitalized in Peking University Shenzhen Hospital from January 2015 to December 2017 were chosen as the subjects.Blood culture and gene chip were both used to detect the pathogens of these infants.The positive rate,the detection time,and the blood volume needed for detection were compared between the two methods.Result A total of 306 cases of suspected neonatal sepsis were included in the study.Among them,34 (11.1%) were positive for blood culture and 54 (17.6%) were positive for gene chip.98 cases were diagnosed as neonatal sepsis,34 (34.7%) were positive for blood culture,and 52 (53.1%) were positive for gene chip.The positive rate of gene chip was higher than that of blood culture (P < 0.05).For the 5 common pathogens of neonatal sepsis,the positive rate of gene chip was higher than that of blood culture.Time to positivity (TTP) and pathogen identification time of blood culture were (14.6 ± 5.5) h and (72.9 ± 19.0) h,respectively.TTP and pathogen identification time of gene chip were both 3 h.The detection time of gene chip was significantly less than that of blood culture (P < 0.001).The blood volume needed for detection of blood culture and gene chip was 1 ~ 2 ml and 0.5 ml.Gene chip needs less blood volume than blood culture.Conclusion Compared with the traditional blood culture,gene chip can quickly detect the pathogens in the blood with higher positive rate and less blood volume.Gene chip is of great value in the diagnosis of neonatal sepsis.

ABSTRACT

The tumor-associated neutrophils (TANs) are thought to be important and complicated character in tumor microenvironment.TANs play dual roles of anti-tumor or tumor-promoting in tumor microenvironment under different factors.TANs can not only play the anti-tumor effect by killing tumor cells directly and promoting immune response,but also can act as tumor promotors by promoting tumor angiogenesis and mediating tumor invasion and metastasis.Recent researches find that TANs are closely related to pre-metastatic niche (PMN),which can promote the construction and maintenance of PMN by means of changing the content of cell factors like interferon-γ.

ABSTRACT

Objective To study the effects and mechanism of different administrations of atorvastatin on myocardial ischemia/reperfusion (MI/R) in rats.Methods A total of 160 male SD rats were randomly divided into five groups:sham group,MI/R group,atorvastatin of conventional dose (MI/R + N) group,atorvastatin of preoperative signal loading dose (MI/R+SL) group,and atorvastatin of preoperative continuous loading dose (MI/ R+ML) group.MI/R model was established in the rats.Myocardial infarction size was detected by Evans blue/ TTC staining.The activity of ATPase of cardiac muscle and the levels of serum IL-6 and TNF-α were detected by ELISA.The level of LVEF% was detected by small animal ultrasound.Results Compared with MI/R+N group,MI/R+ SL and MI/R+ ML groups had significantly smaller myocardial infarction size (P<0.05),higher activity of ATPase (P<0.05),lower levels of serum IL-6 andTNF-α (P<0.05),and more advancedLVEF% (P<0.05).However,MI/R+SL group and MI/R+ML group did not differ significantly in the above-mentioned parameters.Conclusion Atorvastatin of loading dose might alleviate MI/R injury by improving ATP metabolism of cardiac muscle and reducing abnormal expressions of inflammation factors.Meanwhile,the administration of preoperative continuous loading dose and preoperative signal loading dose of atorvastatin may not differ in protecting against MI/R injury.

ABSTRACT

Objective To explore the use of Swedish medical company CMS 4.6.4 radiotherapy XIO system for the design process of cervical cancer postoperative IMRT plans in different iterative times of the same case,and to observe the Segment Weight Optimization(SWO)effect generated after the plan results.Methods Ten cases of cervical cancer patients were chosen ,the use of XIO 4.6.4 CMS treatment system was performed for the development of the intensity modulated radiation therapy .In the SWO process , iterative times were used to generate different plans ,without affecting the clinical dose target area requirements , and compared the organs at risk( OAR) by determining whether there were differences between dose and the number of segments ,monitor units (MU).Results For the same patients,different iterative times within a certain range of SWO IMRT plan were selected to compare the different iterative times plan ,femoral head,rectum and the bladder dose did not change the basic plan.The total number of segments and MU did not change significantly (P>0.05).Conclusion IM-RT plans in the use of CMS XIO 4.6.4 design of cervical cancer after operation in the process of using the itera-tive times below 100 times for SWO is the most suitable method for ensuring the organ dosimetry in radiotherapy . The total number of segments and the MU reduces the radiotherapy plan time so as to improve work efficiency .
